Yes you would need to go into IIS and right click on the Defender Website > Edit Bindings > Add a new HTTPS port > and select the Server's certificate and save the changes then you can delete the HTTP binding.

You can add 2FA to a Local SPP user and select Defender as Radius with secondary authentication, However, you must have an AD account for the username that will be used for the secondary authentication because Defender authenticates users that are based in MS AD where it needs to read\write defender attributes such as token data etc.
